Pharmacological activity arises from agonism at the glucagon-like peptide-1 receptor, a G protein-coupled receptor expressed in the pancreas, the gastrointestinal tract, and the brainstem. Receptor activation raises intracellular cyclic adenosine monophosphate and enhances insulin release in a glucose-dependent manner, an effect that diminishes when blood glucose concentration is low. Other effects include slowed gastric emptying and hypothalamic satiety signalling. These pathways are described well. Receptor desensitisation rates across tissues, relative to the endogenous hormone, are still under investigation, and reported findings differ between laboratories.
The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
Lyophilized peptide material is typically stored at or below -20 °C, with -80 °C used for longer-term archives. Vials should remain sealed and desiccated because moisture promotes aggregation and hydrolysis. Repeated freeze-thaw cycles are avoided since they can alter peptide conformation and reduce recovery. Once reconstituted, solutions are generally kept at 2-8 °C and used within a defined window. Stability beyond those windows depends on buffer composition and concentration, and exact limits are product-specific rather than universal.

The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
There is controversy over GMOs, especially with regard to their release outside laboratory environments. The dispute involves consumers, producers, biotechnology companies, governmental regulators, non-governmental organizations, and scientists. Many of these concerns involve GM crops and whether food produced from them is safe and what impact growing them will have on the environment. These controversies have led to litigation, international trade disputes, and protests, and to restrictive regulation of commercial products in some countries. Most concerns are around the health and environmental effects of GMOs. These include whether they may provoke an allergic reaction, whether the transgenes could transfer to human cells, and whether genes not approved for human consumption could outcross into the food supply.
Some receptor agonists may cause downregulation of their respective receptors, while most receptor antagonists temporarily upregulate their respective receptors. The disequilibrium caused by these changes often causes withdrawal when the long-term use of a drug is discontinued. Upregulation and downregulation can also happen as a response to toxins or hormones. An example of upregulation in pregnancy is hormones that cause cells in the uterus to become more sensitive to oxytocin.

Colombia, officially the Republic of Colombia, is a country located in South America, with insular regions in North America. Colombia's mainland is bordered by the Caribbean Sea to the north, Venezuela to the east, Brazil to the southeast, Peru and Ecuador to the south and southwest, the Pacific Ocean to the west, and Panama to the northwest. Colombia is divided into 32 departments. The Capital District of Bogotá is the country's largest city hosting the main financial and cultural hub. Other urban areas include Medellín, Cali, Barranquilla, Cartagena, Bucaramanga, Pereira, Santa Marta, Cúcuta, Ibagué, Villavicencio and Manizales. It covers an area of 1,141,748 square kilometers (440,831 sq mi) and has a population of around 52 million. Its rich cultural heritage—including language, religion, cuisine, and art—reflects its history as a colony, fusing cultural elements brought by immigration from Europe and the Middle East, with those brought by the African diaspora, as well as with those of Indigenous civilizations that predate colonization. Spanish is the official language, although Creole, English and 64 other languages are recognized regionally. Colombia has been home to many Indigenous peoples and cultures since at least 12,000 BCE. The Spanish landed in La Guajira in 1499, and by the mid-16th century had colonized much of present-day Colombia, establishing the New Kingdom of Granada. Independence from the Spanish Empire was declared in 1810, with what is now Colombia emerging as the United Provinces of New Granada.
=== Streptomyces === Daptomycin is another naturally occurring lipidated peptide, produced by the Gram positive bacterium Streptomyces roseoporous. The structure of Daptomycin consists of a decanoyl lipid chain attached to a partially cyclised peptide head group. It has very potent antimicrobial properties and is used as an antibiotic to treat life-threatening conditions caused by Gram positive bacteria including MRSA (methicillin-resistant Staphylococcus aureus) and vancomycin resistant Enterococci. As with the Bacillus subtilis lipidated peptides, the permeation of the cell membrane is what gives it its properties, and the mechanism of action with daptomycin is thought to involve the insertion of the decanoyl chain into the bacterial membrane to cause disruption. This then causes a serious depolarization resulting in the inhibition of various synthesis processes including those of DNA, protein and RNA, leading to apoptosis.

== Production == The novel was partly inspired by Stephenson's involvement with the Clock of the Long Now project, to which he contributed three pages of sketches and notes. A separate compact disc, entitled IOLET: Music from the World of Anathem, containing eight experimental vocal compositions by David Stutz, was sold separately through CD Baby and the Long Now Foundation, with profits going to The Clock of the Long Now project. To create the world of Arbre, Stephenson constructs new vocabulary. In order to familiarize the reader with the new words, many of which are analogous to English, Latin or Greek words and ideas, a glossary is included at the end of the book. Each chapter begins with a definition of one of these words, which usually relates to the chapter in some way. In addition, the Orth language spoken by the characters was created by Jeremy Bornstein at the author's request, and has been documented. The word anathem was invented by Stephenson, based on the word anthem and the Greek word anathema. In the book, an anathem is a mathic ritual by which one is expelled from the mathic world.
=== 20th century === In 1916, during the First World War, a German Zeppelin airship was shot down during an aerial battle over Billericay. During its fiery demise, it narrowly missed the High Street, crashing into a field off Greens Farm Lane. A plaque was erected at the site in 2016, to commemorate 100 years since the incident. Parts of the aluminium frame can be seen at the Cater Museum in the High Street. Recent research has indicated that this may be identified with the 'ghost Zeppelin' of Tonbridge, which was allegedly seen floating over that town earlier in the day. The former workhouse became St Andrew's Hospital, which housed the internationally renowned Regional Plastic Surgery and Burns Unit from 1973, until this was relocated to Broomfield Hospital, north of Chelmsford, in April 1998. Most of the hospital was redeveloped subsequently into housing, with the listed old workhouse buildings being converted to residential use.
A biosynthesis alternative utilizes genetically engineered yeast species Saccharomyces cerevisiae to synthesize retinal and retinol, using xylose as a starting substrate. This was accomplished by having the yeast first synthesize β-carotene and then the cleaving enzyme β-carotene 15,15'-dioxygenase to yield retinal.
== Soluble TCR == A soluble TCR is a TCR that has been made water-soluble using protein engineering. Such a protein is based on changing an existing TCR's chains to remove the transmembrane portions, leaving only the extracellular part. A number of techniques from protein engineering are then used to stabilize the complex and improve its binding characteristics. A soluble TCR can be used as an antigen-binding module, much like how monoclonal antibodies are used in medicine. Their advantage over antibodies is that they make use of the MHC/HLA display mechanism, allowing them to detect not only extracellular antigens, but also fragments of intracellular ones. On the other hand, they also face some restrictions in terms of applicable HLA genotypes due to them being made to target the complex of a MHC protein with the antigen fragment. One idea for overcoming this restriction is to instead target monomorphic HLA-like molecules such as HLA-E, MHC-related protein 1 (MR1) and cluster of differentiation 1 a, b, c and d (CD1a, CD1b, CD1c or CD1d). An example of a protein derived from soluble TCR is tebentafusp, approved by the US FDA in 2022. This designed protein contains a soluble TCR (αβ) targeting a fragment of gp100 presented by HLA-A*02:01 connected to a ScFv that binds CD3. As a result it causes T cells to activate in the vicinity of cells that present gp100, acting as a molecular glue (a bi-specific T-cell engager in the broad sense). It is only applicable to people with the HLA-A*02:01 genotype.

However, in cases of severe kidney dysfunction the CrCl rate will overestimate the GFR, because hypersecretion of creatinine by the proximal renal tubules will account for a larger fraction of the total creatinine cleared. Ketoacids, cimetidine, and trimethoprim reduce creatinine tubular secretion and therefore increase the accuracy of the GFR estimate, in particular in severe kidney dysfunction. (In the absence of secretion, creatinine behaves like inulin.) An alternative estimation of kidney function can be made when interpreting the blood plasma concentration of creatinine along with that of urea. BUN-to-creatinine ratio (the ratio of blood urea nitrogen to creatinine) can indicate other problems besides those intrinsic to the kidney; for example, a urea concentration raised out of proportion to the creatinine may indicate a prerenal problem, such as volume depletion. Counterintuitively, supporting the observation of higher creatinine production in women than in men, and putting into question the algorithms for GFR that do not distinguish for sex, women have higher muscle protein synthesis and higher muscle protein turnover across their life span. As HDL supports muscle anabolism, higher muscle protein turnover links increased creatine to the generally higher serum HDL in women compared with serum HDL in men.
Lord Robert Cecil on 2 December 1917, assured an audience that the government fully intended that "Judea [was] for the Jews." Yair Auron opines that Cecil, then a deputy Foreign Secretary representing the British Government at a celebratory gathering of the English Zionist Federation, "possibly went beyond his official brief" in saying (he cites Stein) "Our wish is that Arabian countries shall be for the Arabs, Armenia for the Armenians and Judaea for the Jews". The following October Neville Chamberlain, while chairing a Zionist meeting, discussed a "new Jewish State." At the time, Chamberlain was a Member of Parliament for Ladywood, Birmingham; recalling the event in 1939, just after Chamberlain had approved the 1939 White Paper, the Jewish Telegraph Agency noted that the Prime Minister had "experienced a pronounced change of mind in the 21 years intervening" A year later, on the Declaration's second anniversary, General Jan Smuts said that Britain "would redeem her pledge ... and a great Jewish state would ultimately rise." In similar vein, Churchill a few months later stated:
=== Strength === Strong salts or strong electrolyte salts are chemical salts composed of strong electrolytes. These salts dissociate completely or almost completely in water. They are generally odorless and nonvolatile. Most group 1 and 2 metals form strong salts. Strong salts are especially useful when creating conductive compounds as their constituent ions allow for greater conductivity. Weak salts or weak electrolyte salts are composed of weak electrolytes. These salts do not dissociate well in water. They are generally more volatile than strong salts. They may be similar in odor to the acid or base they are derived from. For example, sodium acetate, CH3COONa, smells similar to acetic acid CH3COOH.
HLA-DM (human leukocyte antigen DM) is an intracellular protein involved in the mechanism of antigen presentation on antigen presenting cells (APCs) of the immune system. It does this by assisting in peptide loading of major histocompatibility complex (MHC) class II membrane-bound proteins. HLA-DM is encoded by the genes HLA-DMA and HLA-DMB. HLA-DM is a molecular chaperone that works in lysosomes and endosomes in cells of the immune system. It works in APCs like macrophages, dendritic cells, and B cells by interacting with MHC class II molecules. HLA-DM protects the MHC class II molecules from breaking down, and regulates which proteins or peptides bind to them as well. This regulates how and when a peptide acts as an antigen initiating an immune response. Thus, HLA-DM is necessary for the immune system to respond effectively to a foreign invader. Impairment in HLA-DM function can result in immunodeficiency and autoimmune diseases.
IUPAC and UNESCO were the lead organizations coordinating events for the International Year of Chemistry, which took place in 2011. The International Year of Chemistry was originally proposed by IUPAC at the general assembly in Turin, Italy. This motion was adopted by UNESCO at a meeting in 2008. The main objectives of the International Year of Chemistry were to increase public appreciation of chemistry and gain more interest in the world of chemistry. This event is also being held to encourage young people to get involved and contribute to chemistry. A further reason for this event being held is to honour how chemistry has made improvements to everyone's way of life.

It is a glucagon-like peptide-1 receptor agonist, often grouped with the incretin mimetics. Its backbone is modified from the human hormone to resist enzymatic degradation and to bind albumin. These two features distinguish it from the native peptide.
No. It shares high sequence identity but carries non-natural modifications that alter its stability and clearance. The naturally occurring hormone is broken down within minutes, while the synthetic analogue circulates far longer.
Attachment to serum albumin reduces renal filtration of the peptide and shields it from enzymatic breakdown. The consequence is a much longer interval between administrations than unmodified peptides allow. The precise contribution of binding affinity to overall duration is still being quantified.
The lyophilized powder is dissolved in a suitable solvent, often sterile water or a buffered diluent, with gentle mixing rather than vigorous shaking. Foaming and shear should be avoided because they can promote aggregation. The resulting solution is then stored cold and protected from light.
