forced degradation raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
This page was last updated on 2026-01-02 and is reviewed periodically as new material appears.
Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.
Handling guidance for research quantities calls for single-use aliquots, an inert atmosphere where practical, and avoidance of repeated freeze-thaw cycles that accelerate aggregation. Certificates of analysis typically report purity by peak area, water content, counter-ion identity, and residual solvent levels. In the scientific literature the compound is usually described by its full amino acid sequence, its registry number, or its structural class rather than by any proprietary label. Reporting standards vary between journals, and reviewers increasingly request raw chromatograms alongside tabulated purity figures. Whether current purity thresholds are adequate for every experimental context is debated.
Reversed-phase high-performance liquid chromatography with ultraviolet detection is the dominant approach for peptide purity assessment, usually paired with mass spectrometry to confirm molecular mass and sequence. Peptide mapping by enzymatic digestion and tandem mass spectrometry locates modifications such as deamidation and oxidation. Quantitation in plasma matrices can be performed by LC-MS/MS after solid-phase extraction. Method validation follows general guidance on accuracy, precision, linearity, and limits of detection. Comparability of results between laboratories, when no shared reference standard is available, remains an open question.

Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
William Dale Archerd (May 5, 1912 – October 29, 1977) was an American serial killer who killed at least three people with insulin injections between 1956 and 1966 in Northern California. He was the first to be convicted of using insulin as a murder weapon in the US, and he is suspected in three more cases.
Under the law of July 19, 1871, civil records registered from March 18, 1871, during the Paris Commune, were annulled and rewritten between August 1, 1871, and September 30, 1871, and in the following years. These revised records are referred to as "corrected records" (actes bâtonnés). Only the rewritten records were included in decennial tables, and their dates can differ significantly from the actual events they record. Despite being invalidated by law, the original corrected records remain in the registers and can be searched like any others. At the 12th arrondissement town hall, an accidental fire destroyed birth records from January 1, 1870, to May 25, 1871. These records were reestablished and are classified alongside those reconstructed in the official civil records reconstruction.
== List of nucleosides and corresponding nucleobases == This list does not include modified nucleobases and the corresponding nucleosides Each chemical has a short symbol, useful when the chemical family is clear from the context, and a longer symbol, if further disambiguation is needed. For example, long nucleobase sequences in genomes are usually described by CATG symbols, not Cyt-Ade-Thy-Gua (see Nucleic acid sequence § Notation).
Cysteine-rich proteins (CRP, cysteine-rich peptide or disulphide-rich peptide) are small proteins that contain a large number of cysteines. These cysteines either cross-link to form disulphide bonds, or bind metal ions by chelation, stabilising the protein's tertiary structure. CRPs include a highly conserved secretion peptide signal at the N-terminus and a cysteine-rich region at the C-terminus.
The Viceroyalty of New Granada was established in 1717, then temporarily removed, and then re-established in 1739. Its capital was Santa Fé de Bogotá. This Viceroyalty included some other provinces of northwestern South America that had previously been under the jurisdiction of the Viceroyalties of New Spain or Peru and correspond mainly to today's Venezuela, Ecuador, and Panama. Bogotá became one of the principal administrative centers of the Spanish possessions in the New World, along with Lima and Mexico City, though it remained less developed compared to those two cities in several economic and logistical ways. Great Britain declared war on Spain in 1739, and the city of Cartagena quickly became a top target for the British. A massive British expeditionary force was dispatched to capture the city, but, after achieving initial inroads, devastating outbreaks of disease crippled their numbers, and the British were forced to withdraw. The battle became one of Spain's most decisive victories in the conflict, and secured Spanish dominance in the Caribbean until the Seven Years' War. The 18th-century priest, botanist, and mathematician José Celestino Mutis was delegated by Viceroy Antonio Caballero y Góngora to conduct an inventory of the nature of New Granada. Started in 1783, this became known as the Royal Botanical Expedition to New Granada. It classified plants and wildlife, and founded the first astronomical observatory in the city of Santa Fe de Bogotá. In July 1801 the Prussian scientist Alexander von Humboldt reached Santa Fe de Bogotá where he met with Mutis.

== Epidemiology == The exact incidence is unknown. 20 - 50% of people have some detectable degree of IRP on defecography conducted on healthy volunteers with no symptoms. IRP is a common finding when defecography is carried out to investigate a defecation disorder. For those patients referred for defecography for investigation of obstructed defecation, IRP is detected in 40% of cases. According to another report, 12 to 31% of people with obstructed defecation had IRP. On the other hand, among those who are referred for defecography for investigation of fecal incontinence, IRP is detected in only 10% of cases. Higher grade of IRP is associated with older age. About 90% of cases of IRP occur in females. About 17% of patients with IRP are females who have never given birth. About 10-11% occur in males. In one systematic review including 1301 patients with IRP across 14 studies, 90% were female and 9% were male.
The variation in the 14C/12C ratio in different parts of the carbon exchange reservoir means that a straightforward calculation of the age of a sample based on the amount of 14C it contains will often give an incorrect result. There are several other possible sources of error that need to be considered. The errors are of four general types:
The first long-acting analogue, insulin glargine, was approved in 2000. Insulin aspart, insulin glulisine, and insulin detemir were all approved by 2005. The second wave of insulin analogues, which include insulin degludec and insulin icodec, started in the 2010s. Insulin analogues are on the World Health Organization's List of Essential Medicines.

Reverse-phase high-performance liquid chromatography with ultraviolet detection is the usual choice, with results reported as area percent. Complementary methods such as size-exclusion chromatography and mass spectrometry are needed because a single separation cannot resolve every impurity class. Purity figures are therefore method dependent and should always be read alongside the technique used.
Oxidation mainly affects methionine residues and is promoted by dissolved oxygen, trace transition metals, and prolonged exposure to light. Buffer choice and the presence of antioxidants in a formulation can alter the rate appreciably. Because the products differ in mass by only a few units, mass spectrometry is often required to detect them.
Published data on long-term ambient stability are limited, so the question remains open rather than settled. Short excursions during transport are common in practice, and many suppliers use insulated packaging with cold packs. Where stability data are absent, cold-chain handling with temperature logging is the safer approach.
Purity is commonly expressed as the percentage of the main peak relative to all integrated peaks in a reversed-phase chromatogram. Related substances and counter-ions are reported separately. Values obtained with different detectors are not always directly comparable.
