=== Chemical additives === Chemical additives are small chemical compounds that are added to the crystallization process to increase the yield of crystals. The role of small molecules in protein crystallization had not been well thought of in the early days since they were thought of as contaminants in most case. Small molecules are thought to help if/when they are incorporated as "packing bridges" in the crystal contact interfaces. It is currently not possible (as of 2017) to rationally determine what small molecules to use to improve chances of crystallization.
=== Absorption and distribution === As calciseptine is injected by the snake into its prey along with the rest of its venom, it does not have to pass the protective barrier of the skin and is injected directly into the tissues and/or bloodstream of the prey. No research has been conducted on the toxicokinetics of calciseptine specifically, but general research about snake toxin dynamics is available. Although the toxic peptides are generally small (about 60 amino acids), their size is sufficient to prevent them from crossing epithelial layers like the blood–brain barrier. Bioavailability measurements have been conducted for several snake venoms. For example, cobra venom has been found to have a bioavailability of 41.7% when injected intramuscular, and for other venoms this may even be less than 10%. These values are quite low compared to those of most therapeutic drugs, which usually have a bioavailability of nearly 100% after intramuscular injection. === Pharmacogenetics === Since amitriptyline is primarily metabolized by CYP2D6 and CYP2C19, genetic variations within the genes coding for these enzymes can affect its metabolism, leading to changes in the concentrations of the drug in the body. Increased concentrations of amitriptyline may increase the risk for side effects, including anticholinergic and nervous system adverse effects, while decreased concentrations may reduce the drug's efficacy. Individuals can be categorized into different types of CYP2D6 or CYP2C19 metabolizers depending on which genetic variations they carry. These metabolizer types include poor, intermediate, extensive, and ultrarapid metabolizers. Most individuals (about 77–92%) are extensive metabolizers, and have "normal" metabolism of amitriptyline. Poor and intermediate metabolizers have reduced metabolism of the drug as compared to extensive metabolizers; patients with these metabolizer types may have an increased probability of experiencing side effects. Ultrarapid metabolizers use amitriptyline much faster than extensive metabolizers; patients with this metabolizer type may have a greater chance of experiencing pharmacological failure. The Clinical Pharmacogenetics Implementation Consortium recommends avoiding amitriptyline in patients who are CYP2D6 ultrarapid or poor metabolizers, due to the risk of a lack of efficacy and side effects, respectively. === Extracellular vesicle cross-talk === Recent studies have highlighted the involvement of RAGE (Receptor for Advanced Glycation End-products) in mediating the intercellular communication through extracellular vesicles (EVs), particularly during inflammatory responses. RAGE, known for its interaction with various ligands including advanced glycation end-products (AGEs), plays a key role in the biogenesis and secretion of EVs from stressed or damaged cells. Extracellular vesicles, such as exosomes, are small lipid-bound vesicles that facilitate cell-to-cell communication by transferring molecular cargo including proteins, lipids, and RNAs between cells. Recent evidence suggests that RAGE-associated vesicular pathways contribute to the exacerbation of inflammation by enabling pro-inflammatory signaling between cells. Isinglass finings are widely used as a processing aid in the British brewing industry to accelerate the fining, or clarification, of beer. It is used particularly in the production of cask-conditioned beers, although many cask ales are available which are not fined using isinglass. The finings flocculate the live yeast in the beer into a jelly-like mass, which settles to the bottom of the cask. Left undisturbed, beer will clear naturally; the use of isinglass finings accelerates the process. Isinglass is sometimes used with an auxiliary fining, which further accelerates the process of sedimentation. Non-cask beers that are destined for kegs, cans, or bottles are often pasteurised and filtered. The yeast in these beers tends to settle to the bottom of the storage tank naturally, so the sediment from these beers can often be filtered without using isinglass. However, some breweries still use isinglass finings for non-cask beers, especially when attempting to repair bad batches. Many vegetarians consider beers that are processed with these finings (such as most cask-conditioned ales in the UK) to be unsuitable for vegetarian diets (although acceptable for pescetarians). According to global data in 2018, along with low-calorie beer and gluten-free beer, beers that are acceptable for strict vegetarians are expected to grow in demand in the coming years.
== Music and lyrics == With songs written primarily on the road, the material is darker than Facelift. "We did a lot of soul searching on this album. There's a lot of intense feelings." Cantrell said, "We deal with our daily demons through music. All of the poison that builds up during the day we cleanse when we play". Drug use was front and center as a lyrical theme on the album. Three tracks ("Junkhead", "God Smack", and "Hate to Feel") specifically reference heroin use and its effects. Staley revealed that the album is semi-conceptual and that there are two basic themes in it. The first theme is about "dealing with kind of a personal anguish and turmoil, which turns into drugs to ease that pain, and being confident that that was the answer in a way. Then later on the songs start to slip down closer and closer to hell, and then he figures out that drugs were not, and are not, the way to ease that pain. Basically, it's the whole story of the last three years of my life." Staley described the other theme as being about "painful relationships and involvements with persons." Staley later expressed regret about the lyrical content of some songs on Dirt, explaining, "I wrote about drugs, and I didn't think I was being unsafe or careless by writing about them ...
