Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.

Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
Routine characterisation of the peptide relies on reversed-phase high-performance liquid chromatography, often paired with ultraviolet detection near 214 nanometres. Related substances such as deamidated, oxidised, and truncated sequences elute at characteristic positions and are quantified by area percentage. Electrospray ionisation mass spectrometry confirms the molecular mass and can resolve some closely related variants. Peptide mapping after enzymatic digestion provides sequence-level verification and is useful when a full identity profile is required. Method parameters such as column chemistry, gradient, and mobile-phase pH influence the separation and must be reported alongside results.
Adal's headquarters were again relocated the following century, this time southward to Harar. From this new capital, Adal organised an effective army led by Imam Ahmad ibn Ibrahim al-Ghazi, (Ahmad "Gurey" or "Gran"; both meaning "the left-handed") and his closest top general and brother in law Garad Matan. Imam Ahmed clan is documented to be from the Geri Koombe, a sub clan of Darod. This 16th-century campaign is historically known as the Conquest of Abyssinia (Futuh al-Habash). During the war, Imam Ahmad pioneered the use of cannons supplied by the Ottoman Empire, which he imported through Zeila and deployed against Abyssinian forces and their Portuguese allies led by Cristóvão da Gama.
White Castle also markets its sandwiches in 20 or 30-hamburger boxes, called a Crave Clutch or Crave Case, respectively. The figure of 30 burgers represents the number that can be produced on one of its standard griddles at the same time. A "Crave Crate" is also offered, with the contents being 100 burgers. To celebrate its 100th year in operation, White Castle re-introduced their original burger, called the 1921 Slider. It features an addition of lettuce, tomato, and caramelized onions. Typically normal sliders have 5 holes in the patty while cooking on a griddle of onions and water to steam the sliders to completion. The 1921 is grilled directly on the griddle instead. A variety of White Castle products (mostly frozen) are also sold in grocery stores. Some locations had been cobranded with Church's Chicken until that co-branding arrangement ended around 2010. Around 2012, White Castle experimented with the Laughing Noodle brand that was to share space with White Castle restaurants. The Laughing Noodle concept was discarded a few years later. The Laughing Noodle brand was developed to offer supplemental variety to a White Castle Restaurant. At least one such location was constructed and operated in Sharonville, Ohio. Although White Castle originated in Wichita, Kansas, the city has not had a restaurant since 1938, nor is there a White Castle restaurant in the entire state of Kansas. White Castle is one of the few restaurant chains that does not have a location in its original city.
=== Scaffold === Scaffolds are used to display the heterologous protein on the bacterial cell surface. There are various scaffolds which have been used such as outer membrane proteins, fimbriae/flagella proteins and CPX (circularly permuted OmpX). The CPX scaffold allows peptide fusion at both termini of the scaffold. OMPs are common scaffolds for bacterial display. Proteins can also be displayed on the bacterial cell surface through the use of autotransporters. Autotransporters form part of the type V secretion system. They usually have three domains: leader sequence at the N-terminal; central passenger domain; autotransporter domain at the C-terminal. The heterologous protein is inserted at the passenger domain. Another method of heterologous protein fusion is fusion with fimbriae/flagella, which are filamentous protrusions on the cell surface. There are many fimbriae on mainly Gram-negative bacteria, so displaying proteins on fimbriae is advantageous over some other surface proteins which are less numerous. A disadvantage of using fimbriae is that there is a relatively small insert size limit of 10-30 amino acids.
Under appropriate conditions, an addition reaction can occur between serine or cysteine residues of the target protein and the nitrile group to form imidic acid esters or thioimidates. This mechanism applies to the dipeptidyl peptidase 4 inhibitor vildagliptin used in diabetes mellitus, as well as to saxagliptin. The antibacterial antibiotic cefmetazole also acts as a covalent inhibitor, in this case targeting a bacterial peptidase. The calcium sensitizer levosimendan is presumed to react with the cardiac troponin protein complex. Such a reactive functional group is also referred to as a warhead. In certain cases, nitrile groups exert their effect primarily through steric interactions (i.e., spatial complementarity) by forming van der Waals forces with amino acid residues. This applies to the tyrosine kinase inhibitor bosutinib, which is used in chronic myeloid leukemia. Crystal structures have been reported in which bosutinib is complexed with various tyrosine kinases. Inhibitors of reverse transcriptase, such as Etravirin and Rilpivirin, are used in combination therapies against HIV. The acrylonitrile substructure of rilpivirine penetrates an aromatic cage composed of tyrosine, phenylalanine, and tryptophan, as demonstrated by the corresponding three-dimensional structure published in 2008. The serotonin reuptake inhibitor citalopram, used in the treatment of depression, was the most frequently prescribed psychotropic drug in Germany in 2016, with 290 million defined daily doses.
In May 2023, Ajinomoto signed a letter of intent for a strategic alliance with Solar Foods, a company that developed Solein, a microbial protein feeding on CO2 as a nutrient source, and obtained a marketing license in Singapore. In November 2023, Ajinomoto acquired US-based Forge Biologics for $620 million. In September 2024, Ajinomoto entered into a global strategic partnership with Danone to reduce greenhouse gas emissions from Danone’s raw milk supply chain. In the same year, through joint research with Professor Chinatsu Kasamatsu (Project Professor) of the Institute for SDGs Promotion at Ochanomizu University in Japan, Ajinomoto became the first in the world to develop a new concept of 'electric seasoning,' which adjusts the taste of food using transdermal electrical stimulation. In March 2025, Ajinomoto decided to participate as a partner in the 'Demonstration Survey for the Restoration Model of Degraded Agricultural Land in Brazil,' a project under the Japan-Brazil Green Partnership Initiative promoted by the governments of Japan and Brazil. In April 2025, Ajinomoto was selected by CDP, an international environmental not-for-profit charity, for its "Climate Change A-List" for fiscal 2024. This is the fifth consecutive year that Ajinomoto has been selected for the A-list.

The British implemented a scorched earth policy under which they targeted everything within the controlled areas that could give sustenance to the guerrillas, making it harder for them to survive. As British troops swept the countryside, they systematically destroyed crops, poisoned wells, burned homesteads and farms, and interned Boer and African men, women, children and workers in concentration camps. The British established mounted raiding columns in support of sweeper columns. These were used to rapidly follow and relentlessly harass the Boers to delay them and cut off escape, while the sweeper units caught up. Many of the 90 or so mobile columns formed by the British to participate in such drives were a mixture of British and colonial troops, but they also had a large minority of armed Africans. The number of armed Africans serving with these columns has been estimated at 20,000. The British Army made use of Boer auxiliaries who had been persuaded to change sides and enlist as "National Scouts". Serving under General Andries Cronjé (1849–1923), the National Scouts were despised as joiners but numbered a fifth of the fighting Afrikaners by the end of the War. The British utilised armoured trains to deliver rapid reaction forces much more quickly to incidents (such as Boer attacks on blockhouses and columns) or drop them off ahead of retreating Boer columns.
==== Organorhodium compounds ==== Rhodium is known for its many organometallic derivatives. Rhodium(I) complexes are commonly used as catalysis, with a few being BINAP-Rh(I), DIPAMP-Rh(I), and BDPP-Rh(I). Cyclopentadienyl complexes of rhodium have been investigated as analogues of ferrocene. The parent is rhodocene, which participates in an unusual monomer-dimer equilibrium: Related cyclopentadienyl compounds include the Rh(I) and Rh(III) half-sandwich complexes (C5H5)Rh(CO)2 and Pentamethylcyclopentadienyl rhodium dichloride dimer (C5(CH3)5RhCl2)2. The latter compound is prepared by the reaction of rhodium trichloride trihydrate and pentamethylcyclopentadiene in hot methanol. A related but cationic family of hydrogenation catalysts arise from cyclooctadiene rhodium chloride dimer, Rh2Cl2(C8H12)2. The cyclooctadiene (C8H12) ligands are easily displaced, and this allows the easy introduction of chiral ligands, leading to asymmetric hydrogenations, including the Nobel Prize-winning route to the chiral drug L-DOPA. When treated with sodium borohydride and carbon monoxide, RhCl(P(C6H5)3)3 converts to the pentacoordinate complex RhH(CO)(P(C6H5)3)3, which is used commercially for the hydroformylation of alkenes. Despite its much higher cost, tris(triphenylphosphine)rhodium carbonyl hydride has displaced cheaper cobalt-based catalysts for this application.

Conversely, in rats, blood cholesterol is inversely correlated with cholesterol consumption: the more cholesterol a rat eats the lower the blood cholesterol. During the first seven hours after ingestion of cholesterol, as absorbed fats are being distributed around the body within extracellular water by the various lipoproteins (which transport all fats in the water outside cells), the concentrations increase. Plants make cholesterol in very small amounts. In larger quantities they produce phytosterols, chemically similar substances that compete with cholesterol for reabsorption in the intestinal tract, thus potentially reducing cholesterol reabsorption. When intestinal lining cells absorb phytosterols, in place of cholesterol, they usually excrete the phytosterol molecules back into the GI tract, an important protective mechanism. The intake of naturally occurring phytosterols, which encompass plant sterols and stanols, ranges between ≈200–300 mg/day depending on eating habits. Specially designed vegetarian experimental diets have been produced yielding upwards of 700 mg/day.

Native GLP-1 is a short-lived peptide cleared within one to two minutes by dipeptidyl peptidase-4 and related enzymes. Semaglutide keeps the receptor-binding backbone but adds substitutions and a lipid chain. These changes block the main cleavage site and allow reversible albumin binding, extending the half-life to roughly 165 hours.
Albumin is the most abundant protein in plasma and carries molecules that bear fatty-acid chains. Binding shields the peptide from renal filtration and from peptidases, keeping a circulating reservoir. Slow release from this reservoir produces sustained receptor occupancy and supports infrequent dosing.
The insulinotropic effect is glucose-dependent, meaning secretion increases mainly when glucose is elevated. This property is often described as lowering the chance of hypoglycaemia when the compound is used alone. Other glucose-lowering agents used at the same time can still cause low blood glucose.
Native GLP-1 is degraded within minutes by circulating enzymes. The synthetic version carries substitutions at positions that block enzymatic cleavage, plus a fatty acid side chain that promotes albumin binding. These two changes together extend circulation time from minutes to roughly a week.
