Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.

The distinction between established facts and open questions matters here. That the peptide binds the GLP-1 receptor and stimulates insulin release in a glucose-dependent manner is well documented. How individual variability in receptor density, gastric emptying rate, and gut microbiome composition shapes response remains an active research area. Long-term outcomes beyond five years of continuous use are not yet fully characterized in published trials, and several extension studies are ongoing.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1, a hormone released from intestinal L cells after food intake. The molecule is a 31-amino-acid backbone modified at three positions to resist cleavage by dipeptidyl peptidase-4, the enzyme that degrades native GLP-1 within minutes. A lysine residue at position 26 carries a linker and a C18 fatty diacid, which promotes binding to serum albumin and slows renal clearance. These changes extend the circulating half-life from roughly two minutes to about one week in humans.
Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.
Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
The column(s) in a GC are contained in an oven, the temperature of which is precisely controlled electronically. (When discussing the "temperature of the column", an analyst is technically referring to the temperature of the column oven. The distinction, however, is not important and will not subsequently be made in this article.) The rate at which a sample passes through the column is directly proportional to the temperature of the column. The higher the column temperature, the faster the sample moves through the column. However, the faster a sample moves through the column, the less it interacts with the stationary phase, and the less the analytes are separated. In general, the column temperature is selected to compromise between the length of the analysis and the level of separation. A method which holds the column at the same temperature for the entire analysis is called "isothermal". Most methods, however, increase the column temperature during the analysis, the initial temperature, rate of temperature increase (the temperature "ramp"), and final temperature are called the temperature program. A temperature program allows analytes that elute early in the analysis to separate adequately, while shortening the time it takes for late-eluting analytes to pass through the column.
In 1917, before the revolution, health conditions were significantly behind those of developed countries. As Lenin later noted, "Either the lice will defeat socialism, or socialism will defeat the lice". The Soviet health care system was conceived by the People's Commissariat for Health in 1918. Under the Semashko model, health care was to be controlled by the state and would be provided to its citizens free of charge, a revolutionary concept at the time. Article 42 of the 1977 Soviet Constitution gave all citizens the right to health protection and free access to any health institutions in the USSR. Before Leonid Brezhnev became general secretary, the Soviet healthcare system was held in high esteem by many foreign specialists. This changed, however, from Brezhnev's accession and Mikhail Gorbachev's tenure as leader, during which the health care system was heavily criticized for many basic faults, such as the quality of service and the unevenness in its provision. Minister of Health Yevgeniy Chazov, during the 19th Congress of the Communist Party of the Soviet Union, while highlighting such successes as having the most doctors and hospitals in the world, recognized the system's areas for improvement and felt that billions of rubles were squandered. After the revolution, life expectancy for all age groups went up. These improvements continued into the 1960s when statistics indicated that the life expectancy briefly surpassed that of the United States; life expectancy started to decline in the 1970s, possibly because of alcohol abuse.

, where C = number of C atoms, X = amplitude of the M ion peak, and Y = amplitude of the M +1 ion peak. 13C-enriched compounds are used in the research of metabolic processes by means of mass spectrometry. Such compounds are safe because they are non-radioactive. In addition, 13C is used to quantify proteins (quantitative proteomics). One important application is in stable isotope labeling by amino acids in cell culture (SILAC). 13C-enriched compounds are used in medical diagnostic tests such as the urea breath test. Analysis in these tests is usually of the ratio of 13C to 12C by isotope ratio mass spectrometry. The ratio of 13C to 12C is slightly higher in plants employing C4 carbon fixation than in plants employing C3 carbon fixation. Because the different isotope ratios for the two kinds of plants propagate through the food chain, it is possible to determine whether the principal diet of a human or other animal consists primarily of C3 plants or C4 plants by measuring the isotopic signature of their collagen and other tissues.
== Illegal drugs == An illegal drug trade operates to distribute illegal drugs. The trade of illegal drugs overlaps with trade in contraband of all sorts. Illegal drug distribution does not overlap in obvious ways with the legal trade of legal drugs.

The small council decided to disregard Viserys' declaration of Rhaenyra as his heir, and conspired to crown Prince Aegon (Viserys' eldest son) as the next king of Westeros. Their precedent to justify Aegon's ascension was the Great Council of 101 AC, where Viserys was chosen over Princess Rhaenys on account of sex. His death was finally revealed to the public a week later, when the silent sisters prepared his body for a cremation ceremony traditional in the Targaryen family. Upon the streets learning of Viserys' passing, a coronation ceremony was held where the greens crowned Aegon as the new Lord of the Seven Kingdoms. Aegon's claim was rejected by a furious Rhaenyra, who had a coronation of her own set up by Daemon and the blacks. The coming struggle for power between Aegon and Rhaenyra came to be known as the Dance of the Dragons, a civil war between the greens and blacks.
==== NIH Toolbox ==== In September 2006, the NIH Blueprint for Neuroscience Research started a contract for the NIH Toolbox for the Assessment of Neurological and Behavioral Function to develop a set of state-of-the-art measurement tools to enhance collection of data in large cohort studies. More than 250 scientists from almost 100 academic institutions contributed to its development. In September 2012, the NIH Toolbox was rolled out to the research community. NIH Toolbox assessments are based, where possible, on Item Response Theory and adapted for testing by computer.
Carbohydrate catabolism is the breakdown of carbohydrates into smaller units. Carbohydrates are usually taken into cells after they have been digested into monosaccharides such as glucose and fructose. Once inside, the major route of breakdown is glycolysis, in which glucose is converted into pyruvate. This process generates the energy-conveying molecule NADH from NAD+, and generates ATP from ADP for use in powering many processes within the cell. Pyruvate is an intermediate in several metabolic pathways, but the majority is converted to acetyl-CoA and fed into the citric acid cycle, which enables more ATP production by means of oxidative phosphorylation. This oxidation consumes molecular oxygen and releases water and the waste product carbon dioxide. When oxygen is lacking, or when pyruvate is temporarily produced faster than it can be consumed by the citric acid cycle (as in intense muscular exertion), pyruvate is converted to lactate by the enzyme lactate dehydrogenase, a process that also oxidizes NADH back to NAD+ for re-use in further glycolysis, allowing energy production to continue. The lactate is later converted back to pyruvate for ATP production where energy is needed, or back to glucose in the Cori cycle. An alternative route for glucose breakdown is the pentose phosphate pathway, which produces less energy but supports anabolism (biomolecule synthesis). This pathway reduces the coenzyme NADP+ to NADPH and produces pentose compounds such as ribose 5-phosphate for synthesis of many biomolecules such as nucleotides and aromatic amino acids.
== Pathology == Tuftsin deficiency can be hereditary or can occur following splenectomy, resulting in increased susceptibility to certain diseases e.g.: infected eczematous dermatitis with draining lymph nodes, otitis and sinusitis. Acquired tuftsin deficiency can occur in granulocyte leukemia, when blood neutrophils failed to show stimulation with synthetic tuftsin or with the serum leukokinin. Serum level of tuftsin was minimal or absent.
Biden campaigned heavily for Democrats in the 2010 midterm elections, maintaining an attitude of optimism in the face of predictions of large-scale losses for the party. After big Republican gains in the elections and Emanuel's departure, Biden's past relationships with Republicans in Congress became more important. He led the successful administration effort to gain Senate approval for the New START treaty. In December 2010, Biden's advocacy for a middle ground, followed by his negotiations with Senate minority leader Mitch McConnell, were instrumental in producing the administration's compromise tax package that included a temporary extension of the Bush tax cuts. The package passed as the Tax Relief, Unemployment Insurance Reauthorization, and Job Creation Act of 2010. Obama delegated Biden to lead negotiations with Congress during the 2011 U.S. debt ceiling crisis. Biden's relationship with McConnell brought about the Budget Control Act of 2011 that solved the crisis. Some reports suggest that Biden opposed proceeding with the May 2011 U.S. mission to kill Osama bin Laden, lest failure adversely affect Obama's reelection prospects. In 2012, Biden made the case for Obama's reelection: "Osama bin Laden is dead and General Motors is alive." Obama named Biden to head the Gun Violence Task Force, created to address the causes of school shootings and consider possible gun control measures in the aftermath of the Sandy Hook Elementary School shooting, in December 2012.

However, that 2006 review pre-dates recent data, which, although still too soon to be certain, suggest that the increase in childhood obesity in the US, the UK, and Sweden might be abating.3–5 A British longitudinal study has found that obesity restricted to childhood has minimal influence on adult outcomes at age 30. The study also found that, while obesity that continues into adulthood has little influence on men's outcomes, it makes women less likely to have ever been employed or to currently have a romantic partner. A 2017 National Bureau of Economic Research paper found that childhood obesity in the United States increases medical costs by $1,354 a year (in 2013 dollars).
=== Oxidation === Oxidation can occur both at sulfur, giving a thiophene S-oxide, as well as at the 2,3-double bond, giving the thiophene 2,3-epoxide, followed by subsequent NIH shift rearrangement. Oxidation with trifluoroperacetic acid demonstrates both reaction pathways. The major pathway forms the S-oxide as an intermediate, which undergoes subsequent Diels-Alder-type dimerisation and further oxidation, forming a mixture of sulfoxide and sulfone products with a combined yield of 83% (based on NMR evidence):
Hegseth had announced on 10 October the formation of a new counternarcotics joint task force, to operate in Latin America, the Western hemisphere, and the area of the United States Southern Command (USSOUTHCOM), to be headed by the II Marine Expeditionary Force, intended "to crush the cartels, stop the poison and keep America safe". On 13 November 2025, Hegseth announced the unveiling of Operation Southern Spear, led by Joint Task Force Southern Spear and using a fleet with robotics and autonomous systems to target Latin American drug trafficking, with operations to begin later in November. A US official told a reporter in November that Hegseth's 13 November unveiling was "a formal operation naming for what the Joint Task Force Southern Spear ... and Southcom have already been doing in theater".
=== Uncertainty in absolute isotope ratios === Because many isotopic reference materials are defined relative to one another using the δ notation, there are few constraints on the absolute isotopic ratios of reference materials. For dual-inlet and continuous flow mass spectrometry uncertainty in the raw isotopic ratio is acceptable because samples are often measured through multi-collection and then compared directly with standards, with data in the published literature reported relative to the primary reference materials. In this case the actual measurement is of an isotope ratio and is rapidly converted to a ratio or ratios so the absolute isotope ratio is only minimally important for attaining high-accuracy measurements. However, the uncertainty in the raw isotopic ratio of reference materials is problematic for applications that do not directly measure mass-resolved ion beams. Measurements of isotope ratios through laser spectroscopy or nuclear magnetic resonance are sensitive to the absolute abundance of isotopes and uncertainty in the absolute isotopic ratio of a standard can limit measurement accuracy. It is possible that these techniques will ultimately be used to refine the isotope ratios of reference materials.

It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
The side chain enables reversible binding to albumin in the bloodstream, which protects the peptide from rapid renal clearance. This extends the interval between administrations compared with unmodified GLP-1.
No. It shares much of the native sequence but carries substitutions and an added side chain. These changes increase stability against enzymatic breakdown.
Native GLP-1 is a short-lived peptide cleared within one to two minutes by dipeptidyl peptidase-4 and related enzymes. Semaglutide keeps the receptor-binding backbone but adds substitutions and a lipid chain. These changes block the main cleavage site and allow reversible albumin binding, extending the half-life to roughly 165 hours.
