Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
Receptor activation occurs at GLP-1 receptors distributed across pancreatic islets, the hypothalamus, and the gastrointestinal tract. Binding triggers G protein signaling that raises cyclic AMP and enhances glucose-dependent insulin release. Because the effect depends on prevailing glucose levels, insulin secretion does not rise when blood sugar is already low. Signaling in the brain and gut also influences appetite and gastric emptying, which is why the compound appears in both metabolic and weight-related research literature.
Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.

Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
Routine characterisation of the peptide relies on reversed-phase high-performance liquid chromatography, often paired with ultraviolet detection near 214 nanometres. Related substances such as deamidated, oxidised, and truncated sequences elute at characteristic positions and are quantified by area percentage. Electrospray ionisation mass spectrometry confirms the molecular mass and can resolve some closely related variants. Peptide mapping after enzymatic digestion provides sequence-level verification and is useful when a full identity profile is required. Method parameters such as column chemistry, gradient, and mobile-phase pH influence the separation and must be reported alongside results.
Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
== Medical uses == Dexrazoxane has been used to protect the heart against the cardiotoxic side effects of chemotherapeutic drugs such as anthracyclines, such as daunorubicin or doxorubicin or other chemotherapeutic agents. However, in July 2011 the European Medicines Agency (EMA) released a statement restricting use only in adult patients with cancer who have received > 300 mg/m2 doxorubicin or > 540 mg/m2 epirubicin and general approval for use for cardioprotection. That showed a possibly higher rate of secondary malignancies and acute myelogenous leukemia in pediatric patients treated for different cancers with both dexrazoxane and other chemotherapeutic agents that are associated with secondary malignancies. On 19 July 2017, based on evaluation of the currently available data the European Commission issued an EU-wide legally binding decision to implement the recommendations of the Committee for Medicinal Products for Human Use (CHMP) on dexrazoxane and lifted its 2011-contraindication for primary prevention of anthracycline-induced cardiotoxicity with dexrazoxane in children and adolescents where high doses (≥ 300 mg/m3) of anthracyclines are anticipated. Dexrazoxane was designated by the US FDA as an orphan drug for "prevention of cardiomyopathy for children and adults 0 through 16 years of age treated with anthracyclines". This decision allows virtually all children to receive dexrazoxane starting with the first dose of anthracycline at the discretion of the treating provider.
In 2024, Meta released a collection of large AI models, including Llama 3.1 405B, which was competitive with less open models. Meta's description of Llama as open-source has been disputed due to Llama's software license, which prohibits it from being used for some purposes, and due to Meta not disclosing the origin of the data used to train the models. DeepSeek released their V3 LLM in December 2024, and their R1 reasoning model on 20 January 2025, both as open-weights models under the MIT license. This release made widely known how China had been embracing using and building more open AI systems as a way to reduce reliance on western software and gatekeeping as well as to help give its industries access to higher-powered AI more quickly. Projects based in China have since become more widely used around the world as well as they have closed at least some of the gap with leading proprietary American models. Since the release of OpenAI's proprietary ChatGPT model in late 2022, there have been only a few fully open (weights, data, code, etc.) large language models released. In September 2025, a Swiss consortium added to this short list by releasing a fully open model named Apertus. In December 2025, the Linux Foundation created the Agentic AI Foundation, which assumed control of some open-source agentic AI protocols and other technologies created by OpenAI, Anthropic and Block. Starting in November 2024, Lightricks began releasing the LTX video models as open weights.
=== Average chemical characteristics === On average, healthy humans eliminate 128 g of fresh feces per person per day with a pH value of around 6.6 as indicated by a Fecal pH test. Fresh feces contains around 75% water and the remaining solid fraction is 84–93% organic solids along with some insoluble phosphate salts. These organic solids consist of: 25–54% bacterial biomass, 2–25% protein or nitrogenous matter, 25% carbohydrate or undigested plant matter, and 2–15% fat. Protein and fat come from the colon due to secretion, epithelial shedding, and gut bacterial action. These proportions vary considerably depending on many factors but mainly diet and body weight. The remaining solids are composed of insoluble calcium and iron phosphate salts, intestinal secretions, small amounts of dried epithelial cells, and mucus.

Naturally occurring titanium (22Ti) is composed of five stable isotopes; 46Ti, 47Ti, 48Ti, 49Ti and 50Ti with 48Ti being the most abundant (73.8% natural abundance). Twenty-three radioisotopes have been characterized, with the most stable being 44Ti with a half-life of 59.1 years and 45Ti with a half-life of 184.8 minutes. All of the remaining radioactive isotopes have half-lives that are less than 10 minutes, and the majority of these have half-lives that are less than one second. The isotopes of titanium range from 39Ti to 66Ti. The primary decay mode for isotopes lighter than the stable isotopes is β+ and the primary mode for the heavier ones is β−; the decay products are respectively scandium isotopes and vanadium isotopes. There are two stable isotopes of titanium with an odd number of nucleons, 47Ti and 49Ti, which thus have non-zero nuclear spin of 5/2− and 7/2− (respectively) and are NMR-active.
The interpretation has been described as the "official" United States version of Cold War history. Although it lost its dominance as a mode of historical thought in academic discussions in 1960s, it continues to be influential.
Sarcin-ricin loop substrate binds RTA active site with target adenine stacking against Tyr80 and Tyr123. Arg180 is positioned such that it can protonate N-3 of adenine and break the bond between N-9 of the adenine ring and C-1' of the ribose. Bond cleavage results in an oxycarbonium ion on the ribose, stabilized by Glu177. N-3 protonation of adenine by Arg180 allows deprotonation of a nearby water molecule. Resulting hydroxyl attacks ribose carbonium ion. Depurination of adenine results in a neutral ribose on an intact phosphodiester RNA backbone.
Wood has been a key material in construction since ancient times, so its preservation by coating has received much attention. Efforts to improve the performance of wood coatings continue. Coatings are used to alter tribological properties and wear characteristics. These include anti-friction, wear and scuffing resistance coatings for rolling-element bearings Architectural coatings or paints are used to paint the exteriors and interiors of buildings, often called or external masonry coatings. Clear varnishes and lacquers are generally excluded. Such products are usually designated for specific purposes such as roof coatings, wall paints, or deck finishes. Coatings are eco-friendly building material that increases the efficiency of energy used and reduces impact on human well-being and the environment. The coatings are typically applied with brushes, rollers or sprayers. Wall coatings come in a variety of types, some of which can be applied by amateurs and DIYers without specialized training or equipment. For example, simple paint or primers can often be applied using brushes or rollers, and many people successfully complete such projects themselves. Most masonry surfaces can be treated an exterior wall coating, such as render, pebbledash, stone, stucco or brick.

Merck Millipore was the brand used by Merck Group's (not US-based Merck & Co.) global life science business until 2015 when the company re-branded. It was formed when Merck acquired the Millipore Corporation in 2010. Merck is a supplier to the life science industry. The Millipore Corporation was founded in 1954, and listed among the S&P 500 since the early 1990s, as an international biosciences company which makes micrometer pore-size filters and tests. In 2015, Merck acquired Sigma-Aldrich and merged it with Merck Millipore. In the United States and Canada, the life science business is now known as MilliporeSigma.
=== Bayliss and Starling (1902) === William Bayliss and Ernest Starling, a physiologist and biologist respectively, wanted to see if the nervous system had an impact on the digestive system. From the work of Martin Heidenhain and Claude Bernard, they knew that the pancreas was involved in the secretion of digestive fluids after the passage of food from the stomach to the intestines, which they believed to be due to the nervous system. They cut the nerves to the pancreas in an animal model and discovered that it was not nerve impulses that controlled secretion from the pancreas. It was determined that a factor secreted from the intestines into the bloodstream was stimulating the pancreas to secrete digestive fluids. This was named secretin: a hormone. In 1905, Starling coined the word hormone from the Greek to arouse or excite which he defined as "the chemical messengers which speeding from cell to cell along the blood stream, may coordinate the activities and growth of different parts of the body".
==== Nixon's role reviewed ==== Decades later, a controversial quote attributed to John Ehrlichman, Nixon's domestic policy advisor, claimed that the war on drugs was fabricated to undermine the anti-war movement and African-Americans. In a 2016 Harper's cover story, Ehrlichman, who died in 1999, was quoted from journalist Dan Baum's 1994 interview notes: "... by getting the public to associate the hippies with marijuana and blacks with heroin, and then criminalizing both heavily, we could disrupt those communities. We could arrest their leaders, raid their homes, break up their meetings, and vilify them night after night on the evening news. Did we know we were lying about the drugs? Of course we did." The veracity of the quote was challenged by Ehrlichman's children, and Nixon-era officials. In the end, the increasingly punitive reshaping of US drug policy by later administrations was most responsible for creating some of the conditions Ehrlichman described. In a 2011 commentary, Robert DuPont, Nixon's drug czar, argued that the Comprehensive Drug Abuse Act had represented a degree of drug reform. He noted that the act had rolled back mandatory minimum sentencing and balanced the "long-dominant law enforcement approach to drug policy, known as 'supply reduction'" with an "entirely new and massive commitment to prevention, intervention and treatment, known as 'demand reduction'". Thus, Nixon was not in fact the originator of what came to be called the "war on drugs".

Later in August, Gorbachev resigned as general secretary of the Communist party, and Russian President Boris Yeltsin ordered the seizure of Soviet property. Gorbachev clung to power as the President of the Soviet Union until 25 December 1991, when the USSR dissolved. Fifteen states emerged from the Soviet Union, with by far the largest and most populous one (which also was the founder of the Soviet state with the October Revolution in Petrograd), the Russian Federation, taking full responsibility for all the rights and obligations of the USSR under the Charter of the United Nations, including the financial obligations. As such, Russia assumed the Soviet Union's UN membership and permanent membership on the Security Council, nuclear stockpile and the control over the armed forces. In his 1992 State of the Union Address, US President George H. W. Bush expressed his emotions: "The biggest thing that has happened in the world in my life, in our lives, is this: By the grace of God, America won the Cold War." Bush and Yeltsin met in February 1992, declaring a new era of "friendship and partnership". In January 1993, Bush and Yeltsin agreed to START II, which provided for further nuclear arms reductions on top of the original START treaty.

Native GLP-1 is a short-lived peptide cleared within one to two minutes by dipeptidyl peptidase-4 and related enzymes. Semaglutide keeps the receptor-binding backbone but adds substitutions and a lipid chain. These changes block the main cleavage site and allow reversible albumin binding, extending the half-life to roughly 165 hours.
Albumin is the most abundant protein in plasma and carries molecules that bear fatty-acid chains. Binding shields the peptide from renal filtration and from peptidases, keeping a circulating reservoir. Slow release from this reservoir produces sustained receptor occupancy and supports infrequent dosing.
The insulinotropic effect is glucose-dependent, meaning secretion increases mainly when glucose is elevated. This property is often described as lowering the chance of hypoglycaemia when the compound is used alone. Other glucose-lowering agents used at the same time can still cause low blood glucose.
Native GLP-1 is degraded within minutes by circulating enzymes. The synthetic version carries substitutions at positions that block enzymatic cleavage, plus a fatty acid side chain that promotes albumin binding. These two changes together extend circulation time from minutes to roughly a week.
